Tom Parkes’ 28th minute red card saw the O’s reduced to 10 men, before goals from Matt Bloomfield and Myles Weston sealed victory for the Chairboys.
The U’s head into the game on a run of three wins in six games, including last weekend’s 2-1 victory at Crewe Alexandra.
Leon Legge’s own goal was sandwiched in between two Luke Berry goals to seal victory for Shaun Derry’s side and they currently sit five points outside the play-offs.
Cambridge have suffered nine defeats from their 20 home games to date and have won just six, which have come against Yeovil Town (1-0), Accrington Stanley (2-1), Cheltenham Town (3-1), Crewe Alexandra (2-1), Notts County (4-0) and Newport County (3-2).
