The O’s go into the tie on a three-match unbeaten run following Saturday’s draw at home to AFC Wimbledon.
Ian Hendon’s side took a 10th minute lead when Jay Simpson registered his 15th goal of the season, but were forced to play the final thirty minutes with 10-men after John Marquis was shown a second yellow card. Adebayo Akinfenwa then equalised for the Dons ten minutes from time to claim a share of the spoils.
Scunthorpe arrive in E10 on the back of a 4-0 home defeat against Peterborough United.
The Iron, who are 14th in League One, fell behind on the fourth minute when Conor Washington fired the visitors ahead. Michael Bostwick then doubled the Posh’s advantage from the penalty spot early in the second half before Washington found the net again twice in the final minutes to seal his hat-trick.

Leyton Orient 6-1 Staines Town
The O’s sealed an emphatic victory in the FA Cup first round as they hit non-league side Staines for six.
Orient raced into a 3-0 advantage after only 13 minutes thanks to Ollie Palmer’s brace either side of a Dean Cox strike, though Staines did pull a goal back through former O Darren Purse. Cox then restored the hosts three-goal advantage with a 30-yard free-kick before half time, and the rout was completed late on when John Marquis and Sean Clohessy bagged their first goals for the Club.
Scunthorpe United 2-1 Southend United
Paddy Madden scored twice as Scunthorpe overcame League One rivals Southend in the FA Cup round.
Madden fired the Iron into a 16th minute lead at Glanford Park, but the lead was not to last for long as Ryan Leonard equalised before the break. With 12 minutes remaining Madden was on hand again however as he popped up with the winning goal to set up a second round tie with the O’s.
John Marquis serves a one-match suspension following his dismissal against AFC Wimbledon last weekend. The forward was booked in both halves and as a result will miss out tomorrow.
QPR loanee Cole Kpekawa is cup-tied, which could allow captain Mathieu Baudry to return to the side having come on as a substitute in the draw against Wimbledon.
Meanwhile, Dean Cox underwent knee surgery in the week and Paul McCallum continues his rehabilitation while Connor Essam edges closer to a return.
For Scunthorpe, Neal Bishop misses out after picking up his fifth booking of the season in the loss to Peterborough United last weekend, while Wolves loanee Tommy Rowe is cup tied.
Former O Stephen Dawson remains out injured as does ex-Orient loanee Jamie Ness. They are joined on the treatment table by David Mirfin (hamstring).

Paddy Madden leads the way for the Iron and can be backed at 5/1 to score the first goal, with former Orient striker Darius Henderson priced at 13/2 alongside Luke Williams.
